[18F]P16-129 is a deuterated N-methyl derivative of florbetapir (Amyvid; AV-45), developed as a PET radiotracer for imaging β-amyloid (Aβ) plaques in the brain. It is designed to aid in the diagnosis of Alzheimer's disease by enabling visualization and quantification of amyloid deposits. Deuteration at the N-methyl site was intended to potentially improve pharmacokinetics and imaging performance compared to its non-deuterated analog. Head-to-head clinical studies have shown that [18F]P16-129 has similar binding characteristics and imaging performance as florbetapir f18 ([^18^F]AV45), with no significant benefit from deuteration but offering an alternative for Aβ PET/CT imaging. The drug is being developed by Five Eleven Pharma[1][2][5].
